You can overcome this problem with a security whitelist exception, but it's much easier to download the Flash Player projector application instead. You can then play SDT (or any other downloadable Flash game) on your desktop. Just launch the app (it doesn't need to be installed or registered) and then open Loader.swf inside the Flash Player app. You can also drag-and-drop your Loader.swf file onto the Flash Player window.
If you're just starting out with the SDT Loader, then you may find it convenient to download a "Loader pack" which contains a few pre-configured mods. The core SDT Loader package (created by @ModGuy) is a good starting point for modding project - in which you download many files, test them out, and gradually build up a personalized gameplay experience. But if you have limited time, or if you just want to see what the game is capable of, then a collection such as Slave Bazaar or DrZombi's Sweet Treats might be more useful.
If you're having trouble using the Flash Player application, then sby's Loader Bundle includes a copy of it (along with a "play SDT loader" shortcut which you simply need to double-click). sby's pack is more technical than the ones mentioned above -- it doesn't tell any cool stories, but it does introduce many new features and game mechanics (X-ray vision, breast expansion, etc).
You're probably running the game in a web browser window. Security policies in many browsers will block Flash applications from accessing local files.
