Assassin's Creed: Brotherhood is a brand new game of the series. It has multiplayer mode. You can choose to play as female characters in that mode.

The brothel you first approach, the owner is held hostage and gets her throat cut by the kidnappers.

Bartolomeo's wife is taken prisoner. Upon your rescue attempt, if you're detected, she's executed with a shot to the head.

Desmond stabs Lucy in the stomach while she's frozen in place, presumably killing her at the end of the game.
